Route 29 provided service between Natural Science and Capulet Lane, serving Sarnia Road. Service was only provided when Western University was in session. During exams, no service was provided after 7 PM on weekdays or on weekends.

Before 6 PM from Natural Science, westbound buses operated west on Elgin Drive, south on Western Road, west on Sarnia Road, south on Wonderland Road, west on Beaverbrook Avenue, and south on Capulet Lane to Oxford Street. After 6 PM from Natural Science, westbound buses operated west on Elgin Drive, south on Oxford Drive, and west on Lambton Drive to reach Western Road.

From Capulet Lane and Oxford Street, eastbound buses operated east on Oxford Street, north on Wonderland Road, east on Sarnia Road, north on Western Road, east on Lambton Drive, and north and northwest on Middlesex Drive to Natural Science.
