Route 38 provided service between Masonville Place and the Stoney Creek neighbourhood, serving the Stoneybrook Heights, Stoney Creek and Uplands neighbourhoods.
From Masonville Place, northbound buses operated east on Fanshawe Park Road, north on Hastings Road, northeast on Chambers Avenue, west on Skyline Avenue, north on Bluebell Road, east on Sunningdale Road, south on Adelaide Street, east on Blackwater Road, east on Thistlewood Drive, and north on South Wenige Drive to Sunningdale Road.
From South Wenige Drive and Sunningdale Road, southbound buses operated east on Sunningdale Road, south and northwest on South Wenige Drive, west on Sunningdale Road, south on Bluebell Road, then followed reverse routing to Masonville Place. Trips were interlined with route 39 at Masonville Place.
