From St Catharines Terminal, buses operate northwest on Carlisle Street, south on King Street, southeast on Ontario Street, southwest on St Paul Street, south on Pelham Road, north and west on Louth Street, through SmartCentres plaza, east and north on Benfield Drive, north on Louth Street, east on Fourth Avenue, southeast on Ontario Street, northeast on Church Street, and southeast on Carlisle Street to St Catharines Terminal.
Route 303 may interline with route 302 at St Catharines Terminal.
On evenings and weekends, service is replaced by route 403.

- November 15, 2023-current - Due to the closure of the St Paul Street West CN Rail Bridge, route is diverted to Downtown via Fourth Avenue. Service removed from Chetwood Street, Merigold Street, and McGuire Street.
