From St Catharines Terminal, buses operate northwest on Carlisle Street, northeast on King Street, east on Church Street, northeast on Niagara Street, east on Facer Street, north on Augusta Avenue, northeast on Garnet Street, north on Oblate Street, southwest on Carlton Street, north and northeast on Niagara Street, west on Lakeshore Road, and north on Arthur Street, looping via east on Beachaven Drive, south on Bogart Street, and west on Wildwood Road to Arthur Street, then following the reverse routing to St Catharines Terminal.
Route 307 may interlines with routes 301, 306, or 311 at St Catharines Terminal.
There is no service on evenings and weekends for the majority of the route. Service on the Port Weller loop is replaced by route 408.
