From St Catharines Terminal, buses operate southeast on Academy Street, northeast on St Paul Street, north on Geneva Street, northwest on North Service Road, loop through Fairview Mall, north on Geneva Street, southeast on Wood Street, northeast on Carlton Street, and north on Vine Street to Parnell Road, where buses continue north on Vine Street as route 302. From Vine/Parnell, buses operate south on Vine Street, southwest on Carlton Street, northeast on North Service Road, loop through Fairview Mall, south on Geneva Street, southwest on Church Street, and southeast on Carlisle Street to St Catharines Terminal.
Route 312 interlines with route 302 at Vine/Parnell. It may interline with routes 302 or 303 at St Catharines Terminal.
On evenings and weekends, service on the portion of the route north/east of Fairview Mall is replaced by route 412.
