Route 320 serves the Confederation Heights neighbourhood. It connects to Brock University, Niagara Region Headquarters, and Towpath Terminal.
From Brock University, buses operate east on Sir Isaac Brock Boulevard, east on Sir Isaac Brock Way, south and west on Schmon Parkway, south on Merrittville Highway, east on Decew Road, north on Abbey Drive, west and northeast on Keefer Road, east and south on Summers Drive, east on Confederation Avenue, north on Collier Road, west on Richmond Street, north on Tupper Drive, east on Sullivan Avenue, and north on Towpath Street. Buses in the opposite direction operate north on Towpath Street, west on Albert Street, and south on Pine Street to Sullivan Avenue, then follow the reverse routing to Brock University.
On evenings and weekends, service is replaced by route 421.
