Route 337 serves the Fourth Avenue and Welland Avenue corridors, providing a one-seat journey through Downtown St Catharines. It connects Niagara Health System complex, SmartCentres plaza, Downtown St Catharines, Seaway Industrial Park, and St Catharines Museum & Welland Canals Centre.

From the St Catharines Museum, buses operate north on Welland Canals Parkway, southwest on Eastchester Avenue, north on Cushman Road, west, north, and east on Seapark Drive, north on Cushman Road, southwest on Welland Avenue, southeast on Clark, southwest on Church Street, and southeast on Carlisle Street to St Catharines Terminal. Buses continue northwest on Carlisle Street, southwest on Church Street, northwest on James Street, southwest on Fourth Avenue, and south on First Street Louth, looping through Niagara Health System Complex. Buses continue north on First Street Louth, west on Fourth Avenue, south on Pennachetti Street, west on Pathstone Way, north on Third Street Louth, and east on Fourth Avenue, then following the reverse routing to St Catharines Museum.

On evenings and weekends, service is replaced by route 437.
