This route provides service on Wilson Avenue, York Mills Road, and Ellesmere Road. It also serves the Rexdale neighbourhood and Humber College North Campus operating via Albion Road, Elmhurst Drive, Brookmere Road, Westhumber Boulevard, Martin Grove Road, John Garland Boulevard, and Humber College Boulevard. This route makes limited express stops at major intersections only. Its western terminal is Humber College and the eastern terminal is Scarborough Centre Station for its one branch, 996. This route only operates during the daytime on weekdays.

History
- March 28, 2016 - As part of a reorganization of the branches of route 96 Wilson
, branch 96E was given its own route number and name and is designated as 186 Wilson Rocket.
- May 12, 2019 - With the implementation of TTC's express network, this route was renumbered and renamed to 996 Wilson Express.
- March 23, 2020 - Due to the COVID-19 pandemic and the re-allocation of service to busier routes, service is suspended until further notice.
- November 23, 2020 - In response to increasing system ridership, full service is restored on this route to pre-pandemic levels.
- January 8, 2023 - Service is extended to Ellesmere Station with discontinuation of route 95C service.
- November 19, 2023 - Route is extended further eastbound to to Scarborough Centre Station as part of the Line 3 Bus Replacement Plan.
- January 5, 2025 - New express stops added in both directions on Elmhurst Drive at Albion Road.
Overnight Service
Overnight service is provided by 396 Wilson between Yonge Street and John Garland & Martin Grove, and by 395 York Mills between Yonge Street and Brimley Road. The rest of the route does not have overnight service.
