- September 3, 2006 - Route 401 started service to provide a direct commute to school for students living in the Greensborough and Upper Cornell communities.
- September 4, 2007 - Route 401 was discontinued due to low ridership.

401 Brother Andre School Special via Box Grove is a school bus route operated by York Region Transit in Markham, Ontario, Canada. This is the second and current iteration of route 401.
Route Details
AM: This route follows the same routing as route 14 east from 14th Avenue/McCowan Road to Copper Creek Drive, then goes north on Box Grove Bypass to Church Street, then follows the Cornell Rouge detour routing used on route 18, then west on 16th Avenue to the school.
PM: Reverse AM routing.
History
- February 17, 2009 - Route 401 started service. Originally, students would use route 85 (now route 16) and transfer at the hospital to route 2A (now route 14). However, route 2A buses would usually skip the hospital stop because they were full from students attending Markham District. By having the dedicated bus to complete the full journey, it skipped the transfer at the hospital and saved nearly 40 minutes of travel time for students south of Highway 7.
- September 5, 2017 - The routing is revised to provide service on Kenilworth Gate and Stoney Stanton Road. Buses in the morning operate clockwise while afternoon buses operate counterclockwise.

AM: This route follows the same routing as the discontinued route 41. The route does a first pass of the school at Fincham Avenue before following the route 41 looping. It then travels north on Main Street Markham and east on 16th Avenue to finally terminate at the school.
PM: The route departs the school and follows the discontinued route 41 looping until Raymerville Drive. It then loops via Raymerville Drive, Stone Mason Drive, 16th Avenue, Main Street Markham, and Snider Drive before continuing on Raymerville Drive, McCowan Road, and Highway 7 to terminate at Markville Mall.
