Route 12 mainly serves Edgeley Boulevard between Bass Pro Mills Drive and Langstaff Road, Langstaff Road between Edgeley Boulevard and Ansley Grove Road, Aberdeen Avenue between Ansley Grove Road and Chancellor Drive, and Pine Valley Drive between Chancellor Drive and Steeles Avenue West.
This route serves the Concord, East Woodbridge, Pine Valley Business Park, and Steeles West Industrial Park communities. It also serves Father Bressani Catholic High School and Chancellor Community Centre.
Route 12 connects to Viva Orange at Highway 7.

Historic Branches
History
- September 4, 2001 - Vaughan Transit was merged into YRT in January 2001 and on this date, this route was renumbered from 2 to 12 in order to avoid route number duplication across the York Region Transit network. It ran from Islington Loop along Steeles Avenue West and Pine Valley Drive to loop via Alderson Avenue, Aberdeen Avenue, Ansley Grove Road, and Chancellor Drive.
- June 26, 2005 - Route 12 was rerouted and extended to Vaughan Mills, which had opened nearly a year earlier. The new route went along Steeles Avenue West, Pine Valley Drive, Chancellor Drive, Ansley Grove Road, Langstaff Road, and Edgeley Boulvard to Bass Pro Mills Drive, where it went around the mall parking lot to reach the terminal bays. In this extension, it replaced the northern part of Vaughan Transit/YRT's route 6, which had been replaced in the southern end by TTC route 37D soon after amalgamation.
- November 6, 2008 - Route 12 was indirectly affected by the York University strike between November 6, 2008 and January 29, 2009 due to a high proportion of riders living along the route who commute to York University.
- February 15, 2009 - Extension of a limited-service branch onto Four Valley Drive to provide a shorter walk for employees.
- December 2012 - Route 12 was declared fully accessible.
- April 19, 2015 - Service after 10pm on weekdays and Saturdays is eliminated.
- September 6, 2020 - Due to low ridership, route is reduced to operate during weekday rush hours with weekday off-peak and Saturday service cancelled. Connections to Father Bressani Catholic High School commenced (school tripper runs added, later cut back after two months to a lone midday run due to low ridership). The Four Valley Drive loop was also removed. School tripper service suspended after April 9, 2021 as all classes switched to online learning.
- September 8, 2021 - The midday school tripper run is replaced with the new route 469.
