Route 4 serves Major Mackenzie Drive between Pine Valley Drive and Woodbine Avenue.
The route serves the Vellore Village, Concord, Maple, Patterson, and Cachet communities. It also serve Cortellucci Vaughan Hospital, Maple High School, Canada's Wonderland, City of Vaughan Municipal Offices, Maple GO Station, Mackenzie Richmond Hill Hospital, Alexander Mackenzie High School, Richmond Hill GO Station, and Bayview Secondary School. One trip in the morning and one in the afternoon also serve St Joan of Arc Catholic High School.

History
Major Mackenzie Drive was originally served by two routes. Vaughan Transit's route 4, ran from Bayview Avenue and Major Mackenzie Drive to Jane Street, looping via Cranston Park Avenue, with a rush hour extension south on Jane Street to Langstaff Road. Richmond Hill Transit also had a route 7, operating during rush hours from Bathurst Street to Woodbine Avenue, looping via Hillmount Road and Markland Street.
- 2001 - York Region Transit took over both services in 2001, and combined the two routes together, with route 4 operating between Jane/Langstaff via Cranston Park to Bayview/Steeles bus loop, and route 4A during rush hours providing extra service between Bathurst and Spadina. This provided service along half of Jane Street, with the southern half provided by 35D Jane. Service to the Jane/Langstaff area was rush-hour only, and service to the Cranston Park area was provided in PM rush hour only during westbound trips.
- May 12, 2002 - Routing is extended in the east to the Woodbine and Hillmount area looping clockwise via Woodbine Avenue, Hillmount Road, and Markland Street to replace route 87. Also, new full-time service was introduced to the Jane/Langstaff and Cranston Park area.
- July 15, 2002 - The eastern-end looping is slightly modified to loop counterclockwise in order to improve service for customers within the industrial area.
- May 11, 2003 - Weekend and holiday service after 8:00 PM starts to short turn at Bayview Avenue.
- March 28, 2004 - The eastern-end looping is further modified to loop via south on Woodbine Avenue, west on Hillmount Road, north on Markland Street, and south and west on Hillmount Road to Woodbine Avenue.
- November 2004 - Vaughan Mills opened, with buses being routed through the terminal in both directions. Route 20 Jane was also introduced at this time, providing through service along Jane Street.
- June 26, 2005 - Route 4 was cut back to loop via Jane Street, Springside Road, and Melville Avenue, with service along Jane Street replaced with route 20. Route 4A was also restructured. The loops at Bathurst and Spadina were eliminated, with the route extended west to Weston Road and east to Angus Glen Community Centre.
- November 20, 2005 - The eastern terminus was later extended east again to Mount Joy GO Station.
- November 16, 2008 - Midday service was introduced on route 4A, and rush hour service was again extended east to Markham-Stouffville Hospital via Donald Cousens Parkway until Cornell Terminal is open.
- April 26, 2009 - Service on route 4 was extended to Vaughan Mills again, providing two-way service along Jane Street. Buses no longer enter the Canada's Wonderland Terminal and instead serve the park via the on-street stops on Jane Street. Service along Springside Road and Melville Avenue is removed.
- September 2010 - Routes 4 and 4A were declared fully accessible.
- June 30, 2013 - Route 4 restructured in order to reinforce the grid network. Service east of Woodbine Avenue was replaced by new route 25, and service along McNaughton Road, Melville Avenue, and Brandon Gate Drive was replaced by new route 26. Route 4A expanded to provide daily service between Weston Road and Woodbine Avenue, using the same looping as route 4.
- September 6, 2015 - With development in the Vellore Village community spreading westward, route 4A was extended westward from Weston Road to Pine Valley Drive.
- November 6, 2022 - Service on the 4 branch along Jane Street to Vaughan Mills is discontinued, with all service operating to Pine Valley Drive. Branch 4A renumbered to 4. Service in both directions divert into the new Major Mackenzie West Terminal.
