This east-west route runs between Bellevue Transit Center and Avondale via Kirkland and Redmond. Weekday service runs from 5:15 AM to 12:30 AM, Saturday service runs from 6:20 AM to 11:30 PM and Sunday service runs from 6:20 AM to 11:30 PM.

Eastbound trips depart Bellevue Transit Center north on 108th Avenue NE, east on NE 10th Street, north on 116th Avenue NE, west on Northup Way and north on 108th Avenue NE to South Kirkland Park & Ride. The route then heads west on NE 38th Pl, north on Lake Washington Blvd NE, north on Lakeview Drive, east on NE 68th Street, north on State Street and north on 3rd Street to Kirkland Transit Center. The route then heads east on Central Way, east on NE 85th Street, east on Redmond Way, north on 161st Avenue NE and east on NE 83rd Street to Redmond Transit Center. The trip then continues east on NE 83rd Street, south on 164th Avenue NE, east on Redmond Way, east on Avondale Way, east on NE Union Hill Road and south on 178th Pl NE to Bear Creek Park & Ride. The route then heads north on 178th Pl NE, west on NE Union Hill Road and north on Avondale Road NE to Avondale Pl NE.

Westbound trips depart Avondale Road NE & Avondale Pl NE south on Avondale Road NE, east on NE Union Hill Road and south on 178th Pl NE to Bear Creek Park & Ride. The route then heads north on 178th Pl NE, west on NE Union Hill Road, west on Avondale Way, west on Redmond Way, north on 164th Avenue NE and west on NE 83rd Street to Redmond Transit Center. The route then continues west on NE 83rd Street, south on 161st Avenue NE, west on Redmond Way, west on NE 85th Street, west on Central Way and south on 3rd Street to Kirkland Transit Center. The route then heads south on 3rd Street, south on State Street, west on NE 68th Street, south on Lakeview Drive, south on Lake Washington Blvd NE, east on Northup Way and north on 108th Avenue NE to South Kirkland Park & Ride. The route then heads south on 108th Avenue NE, east on Northup Way, south on 116th Avenue NE, west on NE 10th Street, south on 110th Avenue NE, east on NE 8th Street, south on 112th Avenue NE and west on NE 6th Street to Bellevue Transit Center.

On weekdays during midday and early evening times, trips on the 250 alternate between terminating at Avondale (and skipping Bear Creek Park & Ride), and terminating at Bear Creek Park & Ride (and skipping Avondale). This provides service approximately every 30 minutes on each branch during these times. The route remains the same between Bellevue Transit Center and Avondale Way and NE Union Hill Road - there is service approximately every 15 minutes on this portion during these times.
