This route services Downsview Park, Centennial College Downsview Campus, and the Stanley Greene neighbourhood via Carl Hall Road, John Drury Drive, Keele Street, and Downsview Park Boulevard. Its eastern terminal is Stanley Greene and the western terminal is Downsview Park Station for its one branch, 101.

History
- June 24, 2007 - New seasonal service begins on this route providing service between Downsview Park and the Toronto Aerospace Museum with Downsview Station. Service will only operate during the midday on weekdays and during the daytime on weekends.
- May 11, 2008 - Service is made permanent and operates year round.
- August 31, 2008 - Bike rack service is added to route 101.
- September 6, 2011 - Due to system-wide service cuts, weekday service between September and April is eliminated. Service will only operate during weekends and Holidays.
- May 9, 2016 - All year weekday service is restored. Buses now operate seven days a week.
- December 21, 2016 - Due to construction on the new Centennial College campus, buses follow a one-way looping via east on Carl Hall Road, east and north on Internal Roadway, north and east on Carl Hall Road, and northeast on Chesswood Drive to Sheppard Avenue West. This routing change is scheduled to continue until December 2018.
- September 3, 2019 - Service is increased during the rush hour on weekdays to accommodate customer demand from the new Centennial College Downsview Campus.
- January 8, 2020 - Until further notice, the routing is changed to temporarily operate continuously along Carl Hall Road. Service is removed from the Internal Roadway.
- September 6, 2020 - The temporary route change to operate continuously along Carl Hall Road is made permanent to provide more direct service to the Centennial College Downsview campus. Buses will no longer loop using the Internal Roadway and will continue to follow the one-way looping through the Downsview Park grounds.
- March 28, 2021 - This route was transferred to Wilson upon the opening of the brand new McNicoll Division.
- March 26, 2023 - Service is extended south via Keele Street looping around Frederick Tisdale Drive, Locust Ridge Gardens, and Stanley Greene Boulevard. Service to Sheppard West Station is discontinued.
