This route serves several local roads in the Flemingdon Park community including Wynford Drive, Concorde Place, St. Dennis Drive, Deauville Lane, and Grenoble Drive. It also provides service along Overlea Boulevard and on Broadview Avenue between Danforth Avenue and O'Connor Drive. The southern terminal is Broadview Station and the northern terminal is Don Mills Road & Wynford Drive for its one branch, 100A.

History
- June 22, 2014 - Due to a system-wide reorganization of route branch designations, branch 100 to Eglinton Station is changed to 100C and branch 100D to Don Mills & Wynford via Linkwood is changed to 100B.
- June 21, 2015 - Route is added to the Ten Minute Network. With service increases during the weekday late evening period, combined service between Broadview Station and Don Mills & Wynford will operate at intervals of ten minutes or better whenever the subway is open.
- September 6, 2015 - Ten minute service is extended to Eglinton Station.
- September 6, 2016 - Branches 100B to Don Mills & Wynford via Linkwood and 100C to Eglinton Station have been temporarily replaced by the new 34C Eglinton East
branch due to Eglinton Crosstown construction.
- June 18, 2023 - Service is temporarily re-routed to Pape Station via Pape Avenue due to track construction at Broadview Station. Buses interline with route 72A at Pape Station.
- November 19, 2023 - Due to delays in the reopening of Broadview Station, the route will continue to operate to Pape Station via Pape Avenue, but the route is no longer interlined with 72A Pape.
- January 7, 2024 - With the reopening of the Broadview Station bus terminal, buses now terminate at Broadview Station and no longer divert to Pape Station.
Overnight Service
Overnight service is provided by 325 Don Mills on Pape Avenue and Overlea Boulevard between O'Connor Drive and Don Mills Road. The rest of the route does not have overnight service, however, the route operates until 1:00 AM every day.
