Route Details
This route provides service on Parliament Street between Bloor Street East and Queens Quay East, and on Queens Quay East between Lake Shore Boulevard East and Dockside Drive. Its northern terminal is Castle Frank Station and the southern terminal is Queens Quay and Sherbourne for its one branch, 65.

History
- July 23, 2000 - With the elimination of the 121 Front-Esplanade route, service on branch 65A is extended to serve Union Station via The Esplanade and Front Street. Service will only operate during the daytime on Saturdays with service along the Esplanade to Union Station being provided by the 72A Pape branch at all other times.
- October 15, 2000 - New Sunday and Holiday daytime service is introduced on branch 65 to The Esplanade.
- June 28, 2003 - New seasonal branch 65D to Union Station via Mill Street is introduced providing service to the Distillery District via Eastern Avenue, Cherry Street, and Mill Street. This new branch will replace service on branch 65A during the daytime on Saturdays. Branch 65A to Union Station via The Esplanade will only operate during the early morning on Saturdays.
- September 6, 2003 - Seasonal branch 65D to Union Station via Mill Street ends. This seasonal branch is replaced next year with the new 172 Cherry Street route.
- May 15, 2004 - Branch 65A daytime Saturday service is replaced by the new 172 Cherry Street route.
- November 23, 2008 - Moved to Birchmount Division. Service extended to 1:00am every day.
- March 29, 2009 - Branch 65A to Union Station is removed due to a duplication of service with 72A Pape.
- October 18, 2021 - Due to the conversion of The Esplanade into a one-way westbound only street between Berkeley Street and Princess Street, buses now loop clockwise via Berkeley Street, The Esplanade, and Princess Street.
- September 4, 2022 - Southern terminus is extended to Dockside Drive via Queens Quay East. The loop along The Esplanade, Princess Street and Front Street is discontinued.
