General
Route 28 operates between Trillium Hospital Bus Terminal and City Centre Transit Terminal. It also serves Cooksville GO Station.
The route primarily serves Confederation Parkway, as well as Fairview Road West, Redmond Road and Central Parkway West. It provides service to the Cooksville, Fairview and City Centre neighborhoods of Mississauga.

Timeline
- September 2, 2003 – Route designated as accessible, with low floor buses used at all times.
- September 4, 2007 – Routing in the City Centre area revised to use Confederation Parkway to Rathburn Road, no longer serving Duke of York and Webb Drive.
- October 26, 2015 - Route revised to provide service to Cooksville GO Station during weekday rush hour periods as a result of elimination of route 62. Weekday rush hour frequency increased to 12 minutes from 15 minutes; weekday midday frequency increased to 15 minutes from 24 minutes.
