The current routing of route 2.

Route 2 mainly serves John Street, Esna Park Drive, and Denison Street.

This route serves the Thornhill, Milliken, Middlefield, and Cedarwood communities. On weekends, it also serves the Rouge Fairways, Legacy, Box Grove, and Cedar Grove communities. It also serves Thornhill Community Centre. One trip in the morning and one in the afternoon also serve Middlefield Collegiate Institute during school times.

Route 2 connects to Viva Blue and Viva Pink at Finch Station and Yonge/Steeles, as well as Viva Green at Denison/Warden during rush hours. A small portion of the route is shared with Viva Blue and Viva Pink, between Finch Station and Yonge/Meadowview.

Route 2 was originally operated by Markham Transit as its South Trunk route. It was renamed 2 Milliken after amalgamation in 2001.

• May 12, 2002 - Route 2A is extended in the east from the Legacy/Boxwood area to Markham-Stouffville Hospital via Ninth Line, and in the west from Alden Road and Esna Park Drive to Woodbine Avenue. Service hours on branch 2A are expanded until 9pm on weekdays.

• May 9, 2004 - Weekday rush hour service is added to the Fairtree development at Markham Road and Steeles Ave.

• September 2007 - The eastern looping was expanded slightly during rush hours to provide service to Eastvale Dr., with all weekday service using it.

• January 27, 2008 - The eastern terminus of route 2A is revised to loop on-street instead of terminating within the Markham-Stouffville Hospital. Buses now loop via east on Highway 7, north on Bur Oak Avenue, and west on Church Street to Ninth Line.

• April 27, 2008 - Limited service into Box Grove on route 2A is expanded. All weekday peak-period trips serve the Box Grove neighbourhood instead of four trips a day.

• June 29, 2008 - Service on route 2 is revised in the east to operate via east on Denison Street, and southwest on Elson Street to Eastvale Drive.

• September 2008 - The eastern loop was expanded via Denison St. and Elson St.

• November 2008 - Route 2 was indirectly affected by the York University strike between November 6, 2008 and January 29, 2009 due to a high proportion of riders living along the route who commute to York University.

• September 2009 - The rush hour diversions to Milliken GO Station meeting trains were replaced by the new route 203 Milliken GO Shuttle.

• December 20, 2009 - The eastern-end looping of route 2A is expanded to serve the Grand Cornell Community. Eastbound buses operate via east on Kenilworth Gate, north on Stoney Stanton Road, and east on Highway 7 to route.

• December 2012 - Route 2 was declared fully accessible.

• June 30, 2013 - Route 2A is renamed and renumbered to route 14 '14th Avenue'. Route 2 was extended east on weekends from Russell Jarvis to the Box Grove Smart!Centres.

• September 4, 2022 - With the opening of the Cornell Terminal, routing is adjusted to serve the terminal, operating as the new eastern terminus after buses serve Markham-Stouffville Hospital. Eastern loop routing along 14th Avenue and Box Grove By-Pass is discontinued.
